Criteria for Choosing a Cloning Vector Insert Size
For projects in which it is desired that a
particular piece of DNA be cloned, one consideration is the size of the insert
DNA. Most general cloning plasmids can carry a DNA insert up to around 15 kb in
size. Inserts in excess of this place constraints on proper replication of the
plasmids (particularly for high-copy-number vectors) and can cause problems
with insert stability. Several types of vectors are available for cloning large
fragments of DNA. These are most commonly used to construct libraries of clones
that are often representative of entire genomes. Library clones are then
screened to identify the particular clone that carries the DNA of interest.
